1. Use a static home page, a custom page template & query_posts() or 2. Edit index.php, add your static text before the Loop and then use query_posts to limit the posts on the first page to 1.

Try resetting your custom permalinks back to the default setting via Settings -> Permalinks and renaming your root .htaccess file. If this works, then set a custom permalink structure back up again.
